การพัฒนาชุดตรวจวิเคราะห์การปนเปื้อนของเชื้อรา Aspergillus สายพันธุ์สร้าง Aflatoxin ในขมิ้นชันและขิง

     จากการพัฒนา การประเมินความเฉพาะเจาะจงและประสิทธิภาพความไวของเทคนิค Loop Mediated Isothermal amplification (LAMP) เพื่อตรวจวิเคราะห์เชื้อรา Aspergillus  สายพันธุ์สร้าง Aflatoxin  ในผลิตภัณฑ์สมุนไพรขมิ้นชันและขิง พบว่าเทคนิค LAMP อาศัยไพรเมอร์จำนวน 2 คู่ที่จำเพาะต่อยีนสร้างอะฟลาทอกซิน 6 บริเวณและเพิ่มปริมาณได้ด้วยอุณหภูมิ 63 องศาเซลเซียสเพียงอุณหภูมิเดียว  ภายในเวลาไม่เกิน 1 ชั่วโมง  เทคนิคนี้มีประสิทธิภาพความไวที่ความเข้มข้นเจือจาง 10^-7 เท่า  มีความเฉพาะเจาะจงกับเชื้อรา Aspergillus สายพันธุ์สร้างอะฟลาทอกซิน จำนวน 100 ตัวอย่าง เช่นเดียวกับเทคนิค ELISA  และเมื่อนำเทคนิค LAMP ไปตรวจวิเคราะห์หาเชื้อรา Aspergillus  สายพันธุ์สร้าง Aflatoxin  ในผลิตภัณฑ์สมุนไพรขมิ้นชันและขิงพบว่า ผลิตภัณฑ์ขิงจำนวน 20 ตัวอย่าง  ตรวจพบเชื้อรา Aspergillus  สายพันธุ์สร้าง Aflatoxin  จำนวน 4 ตัวอย่าง ส่วนผลิตภัณฑ์ขมิ้นชันจำนวน 6 ตัวอย่าง ตรวจพบเชื้อราจำนวน 3 ตัวอย่าง  ดังนั้นวิธี   Loop Mediated Isothermal amplification (LAMP)สามารถนำมาประยุกต์ใช้เพื่อตรวจหาเชื้อรา Aspergillus  สายพันธุ์สร้าง Aflatoxin ได้โดยมีความเฉพาะเจาะจงและมีความไวสูง รวมทั้งให้ผลที่รวดเร็วและเชื่อถือได้

     Development of test kit to evaluated the specificity and sensitivity of a Loop Mediated Isothermal amplification (LAMP) method for analyze the contamination of Aspergillus  produce aflatoxin in Curcumin and Ginger products. LAMP technique was designed a pair of outer primers and a pair of inner primers for recognizing six distinct sequences on aflatoxin gene.  Amplifications were observed with sixty three Celsius in one hour.  Sensitivity of this technique was 10^-7 fold and specific with Aspergillus produce aflatoxin as same as ELISA technique.  Determination of Aspergillus produce aflatoxin in Curcumin and Ginger products using LAMP demonstrated that Aspergillus produve aflatoxin was found in three of six Curcumin products and four of twenty Ginger products.  LAMP was proven to be rapid, highly sensitive and specific method for detection of Aspergillus  produce aflatoxin.
